Notch Signaling Pathway03:14

Notch Signaling Pathway

The Notch signaling pathway is a major intracellular signaling pathway that is highly conserved over a broad spectrum of metazoan species. It stands unique from other intracellular signaling mechanisms in animals because notch protein itself acts as the receptor as well as the primary signaling molecule.
The Notch gene came into the limelight in 1914 after the discovery that its mutation in Drosophila melanogaster leads to a serrated (or "notched") wing margin phenotype. It was not until 1985...

Role Of Notch Signalling In Intestinal Stem Cell Renewal01:12

Role Of Notch Signalling In Intestinal Stem Cell Renewal

Notch signaling was first discovered in Drosophila melanogaster, where it is involved in cell lineage differentiation. Notch signaling regulates the maintenance and differentiation of intestinal stem cells or ISCs by controlling the expression of atonal homolog 1 or Atoh1. Atoh1 directs cells to differentiate into secretory cells.
Direct cell-to-cell contact is needed for the activation of Notch signaling. The signal is initiated when a notch ligand binds to a receptor on an adjacent cell, also...
T Cell Activation and Clonal Selection01:22

T Cell Activation and Clonal Selection

T cells are integral to our adaptive immune system, recognizing and effectively responding to foreign antigens. T cell activation and clonal selection are pivotal in orchestrating this immune response. This article elucidates these mechanisms, detailing the roles of cluster of differentiation (CD) markers, major histocompatibility complex (MHC) molecules, costimulatory signals, and the process of clonal selection.
Naive T cells that have not yet encountered an antigen express two primary CD...
Cells of the Adaptive Immune Response01:23

Cells of the Adaptive Immune Response

The T and B lymphocytes of the adaptive immune system develop from common lymphoid progenitor cells in the bone marrow. These progenitors give rise to precursors that eventually develop into both T and B lymphocytes. As these precursors mature, they gain the ability to detect and respond to foreign antigens in the body, a process known as immunocompetence. Additionally, these precursors acquire self-tolerance, a process that ensures they do not react to self-antigens. This intricate system...

Summary

Delta-like 4 signaling through Notch 1 inhibits pro-T cell differentiation into dendritic cells and B cells. Without Notch 1, B cell development in the thymus occurs via a cell-extrinsic pathway.

Area of Science:

  • Immunology
  • Developmental Biology
  • Cell Biology

Background:

  • Pro-T cells are precursors to T lymphocytes.
  • Notch signaling is crucial for cell fate determination.
  • Delta-like 4 (DLL4) is a ligand in the Notch pathway.

Discussion:

  • DLL4 binding to Notch 1 on pro-T cells blocks their differentiation into dendritic cells and B cells.
  • This study reveals a novel regulatory mechanism in early lymphocyte development.
  • The findings highlight the context-dependent role of Notch signaling.

Key Insights:

  • Notch 1 activation by DLL4 restricts T cell lineage commitment.
  • Absence of Notch 1 signaling permits B cell development from thymic precursors.
  • B cell development in this context is regulated by extrinsic factors.
