Summary
The anaphase promoting complex/cyclosome (APC/C) with its adaptor Cdh1 regulates mitosis and prevents premature cell cycle entry. This review summarizes APC/Cdh1 functions in cell cycle control and other cellular processes.
Area of Science:
- Cell Biology
- Molecular Biology
- Biochemistry
Background:
- The anaphase promoting complex/cyclosome (APC/C) is a crucial E3 ubiquitin ligase essential for cell division.
- APC/C utilizes adaptor proteins Cdc20 and Cdh1 to target substrates for degradation.
- Both APCCdc20 and APCCdh1 play vital roles in mitotic progression.
Purpose of the Study:
- To review the multifaceted functions of the APC/Cdh1 complex.
- To explore APC/Cdh1's role in cell cycle regulation, including mitosis and G1/S transition.
- To discuss APC/Cdh1's involvement in non-cell cycle-related cellular processes.
Main Methods:
- Literature review of existing research on APC/C and its adaptors.
- Analysis of studies investigating substrate specificity and regulation of APC/Cdh1.
- Synthesis of findings on APC/Cdh1's impact on cell cycle progression and other cellular functions.
Main Results:
- APCCdh1 mediates the ubiquitination and degradation of key mitotic regulators like cyclin B1, securin, and Plk1.
- APCCdh1 prevents premature S phase entry by controlling mitotic cyclin levels in G1.
- Evidence suggests APCCdh1 regulates cellular processes beyond cell cycle control.
Conclusions:
- APC/Cdh1 is a critical regulator of mitosis and cell cycle progression.
- APCCdh1's functions extend to diverse cellular processes, highlighting its broad biological significance.
- Further research into APC/Cdh1 mechanisms will illuminate its role in both normal physiology and disease states.

Anaphase Promoting Complex
The stepwise destruction of specific proteins is necessary for the progression and completion of the cell cycle. Such proteins are ubiquitinated by ubiquitin ligases and then subsequently destroyed by the proteasome. The SCF (Skp1/Cullin/F-box) and the anaphase-promoting complex (APC) are two important ubiquitin ligases involved in cell cycle progression. While SCF is active throughout the cell cycle, APC gets activated during metaphase to anaphase transition. Cdc20 or Cdh1 binds to APC and...

Inhibition of Cdk Activity
The orderly progression of the cell cycle depends on the activation of Cdk protein by binding to its cyclin partner. However, the cell cycle must be restricted when undergoing abnormal changes. Most cancers correlate to the deregulated cell cycle, and since Cdks are a central component of the cell cycle, Cdk inhibitors are extensively studied to develop anticancer agents. For instance, cyclin D associates with several Cdks, such as Cdk 4/6, to form an active complex. The cyclin D-Cdk4/6 complex...

The Cell Cycle Control System
The cell cycle regulation directs how a cell proceeds from one phase to the next and begins mitosis. The cell cycle control system includes intracellular regulatory molecules and external triggers. They provide "stop" or "advance" signals and operate at specific cell cycle stages termed checkpoints to ensure that a particular process is completed before the cell advances to the next phase.

The Cell Cycle Control System
The cell cycle is an organized set of events that leads the cell to divide into two daughter cells, each containing chromosomes identical to the parent cell. It is the cell cycle that leads to the formation of an entire organism from a single-cell zygote. Besides, cell division also functions in the renewal or repair of tissues in adult multicellular eukaryotes. For example, in the bone marrow, the stem cells divide to form new blood cells. Although essential for several functions, cell...
