A new step nonisothermal experiment efficiently determines drug stability kinetics. This method provides comparable results to traditional isothermal methods but requires significantly less experimental time for captopril oxidation studies.
Area of Science:
- Pharmaceutical Chemistry
- Chemical Kinetics
- Analytical Chemistry
Background:
- Drug stability is crucial for pharmaceutical product efficacy and safety.
- Understanding degradation pathways, such as captopril oxidation, informs formulation and storage.
- Traditional isothermal methods for kinetic studies can be time-consuming.
Purpose of the Study:
- To introduce and validate a novel step nonisothermal experiment for drug stability studies.
- To determine the kinetics parameters of captopril oxidation in aqueous solution.
- To compare the efficiency and precision of the new method with isothermal-isobaric experiments.
Main Methods:
- A step nonisothermal experiment was developed and applied under high oxygen pressure.
- Kinetics parameters for captopril oxidation were determined using this method.
- Computer simulations were employed to statistically evaluate kinetic parameters (E(a) and k(0)).
Main Results:
- The step nonisothermal method yielded comparable kinetic parameters to isothermal-isobaric experiments.
- Aerobic and anaerobic degradation pathways of captopril were distinguished, affecting product formation.
- While isothermal methods offered slightly higher precision, the step nonisothermal method drastically reduced experimental duration.
Conclusions:
- The step nonisothermal experiment is a viable and time-efficient alternative for drug stability kinetic studies.
- This method allows for the determination of rate constants for both aerobic and anaerobic degradation.
- The findings support the optimization of drug stability testing protocols.
