Summary
Cross-linking Fc gamma receptor III (CD16) on monocytes activates pathways that control cytokine release. Phosphoinositide 3-kinase (PI3K) limits tumor necrosis factor-alpha (TNF-alpha) production by inhibiting glycogen synthase kinase-beta (GSK-3beta) and nuclear factor-kappa B (NF-kappaB).
Area of Science:
- Immunology
- Cell Signaling
- Molecular Biology
Background:
- Immunoglobulin G (IgG) complexes engage Fc receptors (FcγRs) like FcγRI (CD64), FcγRII (CD32), and FcγRIII (CD16) on leukocytes.
- Fc receptor engagement activates mitogen-activated protein kinase (MAPK) and phosphoinositide 3-kinase (PI3K) pathways, influencing cytokine production.
Purpose of the Study:
- To elucidate the specific signaling molecules governing cytokine production in monocytes following CD16 cross-linking.
- To investigate the interplay between MAPK, PI3K, glycogen synthase kinase-beta (GSK-3beta), and nuclear factor-kappa B (NF-kappaB) in this process.
Main Methods:
- Monocytes were activated, and cytokine levels (TNF-alpha, IL-1beta, IL-6) were measured after inhibiting MEK1/2, PI3K, and GSK-3beta.
- Reporter assays and small interfering RNA (siRNA) were employed to determine the roles of GSK-3beta and NF-kappaB.
Main Results:
- MAPK signaling stimulated TNF-alpha release, whereas active PI3K limited the production of TNF-alpha, IL-1beta, and IL-6.
- PI3K was found to inhibit NF-kappaB nuclear translocation.
- The inhibitory effect of PI3K on TNF-alpha production was linked to reduced GSK-3beta activity, which in turn decreased NF-kappaB transactivation.
Conclusions:
- CD16 cross-linking activates PI3K in monocytes.
- Active PI3K limits TNF-alpha production by suppressing GSK-3beta activity, thereby inhibiting NF-kappaB-mediated transcription.
- NF-kappaB plays a crucial role in TNF-alpha production induced by CD16 cross-linking.
