Summary

High peptide density on biomaterials doesn't always improve cell adhesion. Flexible linkers like polyethylene oxide (PEO) may enhance cell interactions more effectively than high-density dendrimers.

  • Quantification of peptide grafting density via (125)I radiolabeling.

    • Successful attachment of polymer linkers and peptides confirmed by surface analysis techniques.
    • Dendrimer-modified surfaces achieved twofold greater peptide grafting density compared to PEO-modified surfaces.
    • Vascular endothelial cell adhesion was significantly higher on PEO-modified surfaces, suggesting improved peptide accessibility.

    Conclusions:

    • While dendrimers can achieve higher surface peptide densities, this does not necessarily translate to improved cell adhesion.
    • Flexible linkers like PEO may enhance cell-surface receptor binding by increasing peptide accessibility.
    • Optimizing peptide surface density alone may be insufficient for enhancing biomaterial-cell interactions; linker properties are also critical.
