Discs Large (Dlg) protein and glutamate receptors are found at mammalian neuromuscular junctions (NMJs). This suggests novel glutamatergic mechanisms may operate at these critical nerve-muscle connections.
Area of Science:
- Neuroscience
- Molecular Biology
- Synaptic Plasticity
Background:
- Discs Large (Dlg) is a multidomain protein known to interact with glutamate receptors and potassium channels.
- Dlg has been observed at Drosophila neuromuscular junctions (NMJs) and mammalian central nervous system synapses.
- Dlg also localizes postsynaptically at cholinergic mammalian NMJs.
Purpose of the Study:
- To investigate the presence and localization of glutamate receptor subunits at mammalian NMJs.
- To determine the colocalization patterns of Dlg, glutamate receptors, and potassium channels at mammalian NMJs.
- To explore potential novel glutamatergic mechanisms at mammalian neuromuscular synapses.
Main Methods:
- Immunohistochemistry to detect protein localization.
- Analysis of NMJs in normal mice.
- Examination of alpha-amino-3-hydroxy-5-methylisoxazole-4-proprionate (AMPA) and N-methyl-D-aspartate (NMDA) receptor subunits, glutamate, Dlg, and Kir2 potassium channels.
Main Results:
- AMPA receptor subunits and glutamate are present at the mammalian NMJ.
- AMPA and NMDA receptor subunits colocalize with Dlg postsynaptically at all examined NMJs.
- Kir2 potassium channels also show overlapping localization with Dlg and glutamate receptors.
Conclusions:
- The presence of glutamatergic components at mammalian NMJs suggests a role for glutamate signaling.
- Dlg, glutamate receptors, and potassium channels form a complex at the postsynaptic site of mammalian NMJs.
- These findings indicate novel glutamatergic mechanisms may be involved in mammalian neuromuscular transmission.

Chemical Synapses
Because chemical synapses depend on the release of neurotransmitter molecules from synaptic vesicles to pass on their signal, there is an approximately one millisecond delay between when the axon potential reaches the presynaptic terminal and when the neurotransmitter leads to opening of postsynaptic ion channels. Additionally, this signaling is...
