Poliomyelitis

Poliomyelitis is caused by poliovirus, a small, non-enveloped, positive-sense RNA virus of the Picornaviridae family and Enterovirus genus. Transmission occurs primarily via the fecal-oral route, often through ingestion of contaminated water or food. The virus initially replicates in the oropharynx and intestinal mucosa, particularly in lymphoid tissues such as the tonsils, Peyer’s patches, and regional lymph nodes. Summary

A novel Ebolavirus vaccine candidate, EbolaDeltaVP30, shows promise. This replication-deficient virus protected mice and guinea pigs from lethal Ebolavirus infections, indicating its potential as a safe and effective vaccine platform.

Background:

  • Ebolavirus causes severe hemorrhagic fever with high fatality rates.
  • No licensed vaccines are currently available for Ebolavirus.
  • A replication-deficient Ebolavirus, EbolaDeltaVP30, was previously developed.

Purpose of the Study:

  • To evaluate the vaccine potential of EbolaDeltaVP30.
  • To assess the safety and protective efficacy of EbolaDeltaVP30 in animal models.

Main Methods:

  • Safety was assessed in STAT-1-knockout mice.
  • Protective efficacy was tested in mice and guinea pigs against lethal Ebolavirus challenge.
  • Humoral and cellular immune responses were analyzed.

Main Results:

  • EbolaDeltaVP30 was found to be safe in susceptible mice.
  • Vaccinated mice and guinea pigs were protected from lethal Ebolavirus infection.
  • Protection correlated with high antibody responses to Ebolavirus glycoprotein and CD8+ T-cell activation.
