Summary

Chemokine receptor CCR4 plays a critical role in Cutaneous T cell lymphoma (CTCL) cell migration. This study shows CCR4 mediates CTCL cell chemotaxis and transendothelial migration, crucial for skin infiltration.

  • Mycosis fungoides (MJ) cells exhibited higher CCR4 expression than Sezary syndrome (Hut78) cells.
  • MJ cells showed significant chemotactic migration, integrin activation (CD49d, CD29), and transendothelial migration in response to CCL22.
  • MJ cells demonstrated enhanced transendothelial migration compared to Hut78 cells.

Conclusions:

  • CCR4 plays a critical role in mediating CTCL cell migration.
  • Findings support the multistep cascade model of leukocyte transendothelial migration.
  • CCR4 is a potential therapeutic target for CTCL.
