The CYP2D6*4/*4 genotype, affecting serotonin metabolism, was not linked to a higher risk of depression or anxiety disorders in elderly individuals. This genetic variation does not predispose older adults to these mental health conditions.
Area of Science:
- Pharmacogenetics
- Neuroscience
- Geriatric Psychiatry
Background:
- 5-methoxytryptamine (5-MT), a serotonin precursor, is metabolized by cytochrome P450 2D6 (CYP2D6).
- Individuals with the CYP2D6*4/*4 genotype lack CYP2D6 enzyme activity, potentially leading to altered serotonin levels.
- This genetic profile might influence susceptibility to depression and anxiety, particularly in older populations.
Purpose of the Study:
- To investigate the association between the CYP2D6*4/*4 genotype and the predisposition to depression or anxiety disorders.
- To determine if genetic variations in CYP2D6 influence mental health risks in the elderly population.
Main Methods:
- A cross-sectional study was conducted within the Rotterdam Study, a population-based cohort.
- Participants aged 55 years and older were screened for depression and anxiety disorders.
- Logistic regression analysis examined the association between the CYP2D6*4 polymorphism and the risk of these disorders.
Main Results:
- No significant association was found between the CYP2D6*4/*4 genotype and an increased risk of major depression (OR = 0.85; p = 0.72).
- The CYP2D6 genotype was not associated with minor depression (OR = 1.56; p = 0.28).
- No elevated risk of anxiety disorders was observed in carriers of the CYP2D6*4/*4 genotype (OR = 1.19; p = 0.55).
Conclusions:
- Genetic variation in the CYP2D6 gene, specifically the *4/*4 genotype, is not associated with a predisposition to depression or anxiety disorders.
- These findings suggest that CYP2D6 genotype does not play a significant role in the development of depression or anxiety in the elderly.
