Constitutional Isomers of Alkanes02:18

Constitutional Isomers of Alkanes

Organic compounds of the same molecular formula can have different structural formulas called constitutional isomers, and the phenomenon is known as constitutional isomerism. Alkanes with four or more carbons showing multiple structures with the same molecular formula thereby exhibit constitutional isomerism.
The linear isomer of an alkane is prefixed by the term “n”; hence a linear isomer of pentane is known as n-pentane. Based on the type of branching, some of the branched isomers are given...
Nomenclature of Alkanes02:22

Nomenclature of Alkanes

In the late 19th-century, the number of new chemical compounds discovered increased tremendously. Hence, the necessity arose to develop a naming system for the systematic nomenclature of these newly discovered compounds. IUPAC (International Union for Pure and Applied Chemistry), established in 1919, sets rules for the nomenclature.
The alkane nomenclature considers the length of the carbon chain, the number, and the location of the substituent to arrive at its systematic name. The IUPAC...
Adrenergic Antagonists: Chemistry and Classification of ɑ-Receptor Blockers01:17

Adrenergic Antagonists: Chemistry and Classification of ɑ-Receptor Blockers

Adrenergic antagonists, or sympatholytics, inhibit adrenoceptor activation driven by catecholamines or agonists. Based on their adrenoceptor specificity, adrenergic blockers can be categorized into two primary groups: α-adrenergic blockers (α-blockers) and β-adrenergic blockers (β-blockers). α-blockers interact with α1 and α2 subtypes of α-adrenoceptors.
Nonselective α-blockers: Nonselective α-blockers contain haloalkylamine or imidazoline moieties. Phenoxybenzamine, with a haloalkylamine...
Adrenergic Antagonists: ɑ and β-Receptor Blockers01:31

Adrenergic Antagonists: ɑ and β-Receptor Blockers

Third-generation β-blockers, such as labetalol and carvedilol, represent a significant advancement in managing cardiovascular conditions. Unlike conventional β-blockers, which can induce peripheral vasoconstriction, third-generation drugs block α1 adrenoceptors. This promotes vasodilation through several mechanisms, such as increased nitric oxide production, inhibition of calcium ion entry, opening of potassium ion channels, and antioxidant action. Labetalol, for instance, is clinically...
Local Anesthetics: Chemistry and Structure-Activity Relationship01:30

Local Anesthetics: Chemistry and Structure-Activity Relationship

Local anesthetics (LAs) are drugs that induce a temporary loss of sensation in a limited body area, preventing pain. Cocaine was the first local anesthetic discovered in the late 19th century. Cocaine is a benzoic acid ester obtained from the leaves of coca shrubs and was often used for its psychotropic effects. Cocaine was first isolated in 1860 by Albert Niemann. Sigmund Freud studied the physiological actions of cocaine. Carl Koller later introduced it into clinical practice in 1884 as a...
Adrenergic Receptors: ɑ Subtype01:31

Adrenergic Receptors: ɑ Subtype

Adrenoceptors are classified into α and ꞵ classes based on their potencies to catecholamine agonists. α-adrenoceptors show the following order of catecholamine potency:
Adrenaline ≥ Noradrenaline >> Isoprenaline
α-adrenoceptors are further divided into α1 and α2-adrenoceptors.
α1-Adrenoceptors: These receptors are located postsynaptically on the effector organs and cause constriction of smooth muscle mediated by activation of phospholipase C—inositol-1,4,5-trisphosphate...

Summary

Researchers isolated two new isomeric acylated oleanane-type triterpenoids, racemosol A and isoracemosol A, from Barringtonia racemosa fruits. These compounds, along with three known ones, showed no activity against tested carcinoma cell lines.

Area of Science:

  • Natural Product Chemistry
  • Phytochemistry
  • Organic Chemistry

Background:

  • Barringtonia racemosa is a plant species known for its diverse phytochemical constituents.
  • Triterpenoids, particularly oleanane-type, are a significant class of natural products with various reported bioactivities.
  • Acylated triterpenoids represent a subclass with potential for novel structural and biological properties.

Purpose of the Study:

  • To isolate and characterize novel compounds from the dried fruits of Barringtonia racemosa.
  • To elucidate the structures of new isomeric acylated oleanane-type triterpenoids.
  • To evaluate the cytotoxic activity of the isolated compounds against specific cancer cell lines.

Main Methods:

  • Extraction of plant material using methanol.
  • Isolation of compounds using chromatographic techniques.
  • Structure elucidation employing spectroscopic methods, including 1D and 2D Nuclear Magnetic Resonance (NMR) spectroscopy, and chemical analyses.
  • Cytotoxicity assays against HeLa and P388 D1 carcinoma cell lines.

Main Results:

  • Two new isomeric acylated oleanane-type triterpenoids, named racemosol A (1) and isoracemosol A (2), were successfully isolated.
  • The structures were determined to be 22alpha-acetoxy-3beta,15alpha,16alpha,21beta-tetrahydroxy-28-(2-methylbutyryl)olean-12-ene (1) and 21beta-acetoxy-3beta,15alpha,16alpha,28-tetrahydroxy-22alpha-(2-methylbutyryl)olean-12-ene (2).
  • A total of five compounds, including three known triterpenoids, were isolated and characterized.
  • None of the isolated compounds (1-5) exhibited significant cytotoxic activity against HeLa and P388 D1 carcinoma cell lines.

Conclusions:

  • The study successfully identified and characterized two novel isomeric acylated oleanane-type triterpenoids from Barringtonia racemosa.
  • The findings contribute to the phytochemical knowledge of Barringtonia racemosa.
  • The isolated compounds did not demonstrate cytotoxic effects on the tested cancer cell lines, suggesting limited potential for these specific compounds in cancer therapy against these cell lines.
