Summary

Forkhead box proteins A1 and A2 (Foxa1/2) are crucial for normal bile duct development. Their absence in embryonic liver leads to abnormal bile duct growth by activating IL-6 expression, a key proliferative signal.

Main Results:

  • Deletion of Foxa1/2 in embryonic liver resulted in significant hyperplasia of the biliary tree and abnormal bile duct formation.
  • This abnormal development was partly due to the activation of IL-6 expression, a cholangiocyte proliferative signal.
  • In Foxa1/2-deficient livers, the glucocorticoid receptor failed to bind the IL-6 promoter, leading to enhanced IL-6 expression.

Conclusions:

  • Foxa1/2 are required for normal bile duct development post-liver specification by preventing excessive cholangiocyte proliferation.
  • Foxa1/2 act as terminators of bile duct expansion in the adult liver by inhibiting IL-6 expression.
