In Vivo, Percutaneous, Needle Based, Optical Coherence Tomography of Renal Masses

9.3K
Optical coherence tomography (OCT) is a high resolution imaging technique that allows analysis of tissue specific optical properties providing the means for tissue differentiation. We developed needle based OCT, providing real-time imaging combined with on-the-spot tumor differentiation. This publication describes a method for percutaneous, needle based OCT of renal...

Summary

The tumor suppressor p53 protein

Area of Science:

  • Molecular Biology
  • Cellular Biology
  • Cancer Research

Background:

  • The canonical p53 activation pathway involves stabilization, DNA binding, and transcriptional activation.
  • Emerging evidence suggests increased complexity in each p53 activation step.
  • The classical p53 activation model may not fully explain in vivo mechanisms.

Purpose of the Study:

  • To propose a revised model for p53 activation.
  • To incorporate the role of antirepression in p53's physiological activation.

Main Methods:

  • Review of existing genetic and in vitro studies.
  • Conceptual model development.

Main Results:

  • Classical p53 activation steps are insufficient to explain all in vivo observations.

  • Antirepression, releasing p53 from inhibitors like Mdm2 and MdmX, is crucial.
  • This release is proposed as a key physiological activation step.
  • Conclusions:

    • Antirepression is a critical, previously underappreciated step in p53 activation.
    • A revised model including antirepression better explains p53's in vivo function.
    • Understanding this mechanism is vital for cancer research and therapy.
