Summary

Endocan (endothelial cell-specific molecule-1) is found in high-grade gliomas, including glioblastomas. Its presence in tumor endothelial cells correlates with shorter patient survival, suggesting a role in abnormal tumor vasculature.

  • Glioblastomas (GBMs) exhibit microvascular proliferation and necrosis.
  • Endocan (endothelial cell-specific molecule-1) is a proteoglycan secreted by endothelial cells.
  • Proangiogenic factors upregulate endocan.

Purpose of the Study:

  • To investigate endocan expression in human glioblastoma cell lines and brain tumors.
  • To determine the correlation between endocan expression and glioma grade and patient survival.

Main Methods:

  • In vitro studies using human GBM cell lines (T98G, U118MG).
  • Upregulation of endocan by specific factors (TNF, FGF2, hypoxia).
  • Immunohistochemical analysis of endocan expression in 82 human brain tumors.

Main Results:

  • Endocan is expressed in vitro by endothelial cells and human GBM cell lines.
  • Endocan is detected in hyperplastic endothelial cells and tumor cells of high-grade gliomas, particularly at tumor margins and in palisading cells.
