Decrease of nicotinic receptors in the nigrostriatal system in Parkinson's disease

Nicotine may protect against Parkinson's disease (PD). This study found reduced nicotinic acetylcholine receptors (nAChRs) in the brains of PD patients, but nAChR levels did not correlate with disease severity.

- Two PET scans were utilized: one for dopaminergic function (6-[18F]fluoro-L-DOPA) and another for nAChRs (2-[18F]fluoro-A-85380).
- Regions-of-interest and voxel-based analyses compared 2-[18F]fluoro-A-85380 distribution volumes (DVs) between groups.

- PD patients exhibited a significant decrease in 2-[18F]fluoro-A-85380 DV in the striatum (-10%) and substantia nigra (-14.9%) compared to controls.
- No significant correlation was observed between nAChR DV and clinical PD severity scores.
- No significant correlation was found between nAChR DV and PET-measured striatal dopaminergic function.
Conclusions:
- This in vivo study confirms reduced nAChR availability in the striatum and substantia nigra of PD patients.
- The observed decrease in nAChRs does not correlate with clinical or dopaminergic markers of PD severity in this cohort.
- Further research is needed to elucidate the role of nAChRs in PD pathogenesis and progression.
