Area of Science:

  • Pharmacology
  • Neuroscience
  • Clinical Medicine

Background:

  • Sertraline hydrochloride is a novel naphthylamino compound.
  • It selectively inhibits the neuronal reuptake of serotonin.
  • The drug is available in the UK and under review in the US.

Purpose of the Study:

  • To evaluate the efficacy and safety of sertraline hydrochloride.
  • To characterize the pharmacokinetic profile of sertraline.
  • To compare the adverse effect profile with other antidepressants.

Main Methods:

  • First-order kinetics analysis for plasma elimination.
  • Assessment of protein binding and volume of distribution.
  • Multicenter clinical studies for efficacy in depression and OCD.

Main Results:

  • Sertraline exhibits a plasma elimination half-life of 24-26 hours.
  • High plasma protein binding and a large volume of distribution observed.
  • Demonstrated efficacy in treating depression and obsessive-compulsive disorder.

Conclusions:

  • Sertraline is an effective treatment for depression and OCD.
  • Adverse effects are mainly gastrointestinal (nausea, diarrhea, dyspepsia).
  • The side effect profile is distinct from tricyclic antidepressants but similar to fluoxetine.
