We characterized the JM8 mouse embryonic stem cell line for germline competence. Restoring the agouti gene simplifies breeding for large-scale mouse knockout programs using the C57BL/6 background.
Area of Science:
- Genetics
- Developmental Biology
- Stem Cell Biology
Background:
- Mouse embryonic stem cells (ESCs) are crucial for genetic engineering.
- The C57BL/6N strain is a standard genetic background for mouse models.
- Existing ESC lines may have limitations in breeding schemes.
Purpose of the Study:
- To characterize the germline competence of the JM8 mouse ESC line.
- To modify JM8 cells for simplified breeding by restoring the agouti coat color gene.
- To establish a resource for large-scale mouse knockout programs.
Main Methods:
- Characterization of germline transmission in JM8 ESCs.
- Targeted gene repair to restore the agouti locus in JM8 cells.
- Assessment of breeding efficiency with modified JM8 cells.
Main Results:
- JM8 ESCs demonstrated high germline competence.
- Targeted repair successfully restored the dominant agouti gene in JM8 cells.
- The modified JM8 line facilitates simplified breeding strategies.
Conclusions:
- The characterized JM8 ESC line is suitable for germline transmission.
- Agouti gene restoration in JM8 cells enhances their utility for mouse genetics.
- These modified ESCs provide a valuable resource for C57BL/6N knockout mouse generation.
