Summary

Coenzyme A (CoA) effectively lowers plasma lipids in hyperlipidaemia patients. It enhances fatty-acid oxidation in the liver, reducing triglyceride and VLDL synthesis.

Background:

  • Primary hyperlipoproteinaemias, including phenotype IV (hypertriglyceridaemia) and IIb (combined hyperlipidaemia), are characterized by elevated plasma lipids.
  • Coenzyme A (CoA) plays a crucial role in lipid metabolism, particularly in acylation reactions.

Purpose of the Study:

  • To investigate the lipid-lowering effects of Coenzyme A (CoA) in patients with primary hyperlipoproteinaemias.
  • To elucidate the underlying mechanisms of CoA's influence on plasma lipid levels and hepatic lipid metabolism.

Main Methods:

  • Administration of CoA to animal models on hyperlipidaemic diets.
  • Analysis of plasma lipid levels and hepatic triglyceride/lipid concentrations.
  • Measurement of fatty-acid oxidation (palmitate oxidation) in isolated liver mitochondria and peroxisomes from treated rats.

Main Results:

  • CoA administration reduced plasma lipid levels in hyperlipidaemic animals.
  • Hepatic concentrations of triglycerides and/or lipids were decreased following CoA treatment.
  • Increased palmitate oxidation was observed in liver mitochondria and peroxisomes of CoA-treated rats.

Conclusions:

  • CoA enhances fatty-acid oxidation in the liver, leading to reduced endogenous triglyceride synthesis and VLDL formation.
  • Stimulation of fatty-acid oxidation in extrahepatic tissues may also contribute to increased VLDL catabolism.
  • CoA demonstrates potential as a therapeutic agent for managing hyperlipidaemia.
