Genetic variations in the GPC5 gene are linked to how well patients with multiple sclerosis (MS) respond to interferon-beta treatment. This study confirms the association of GPC5 polymorphisms with treatment outcomes in MS.

Background:

  • Multiple sclerosis (MS) is a chronic central nervous system inflammatory disease.
  • Interferon-beta is a common treatment for relapsing-remitting MS, but efficacy varies significantly among patients.
  • Genome-wide association studies have identified genetic polymorphisms influencing interferon-beta response.

Purpose of the Study:

  • To replicate findings on the association between genetic loci and interferon-beta treatment response in MS patients.
  • Specifically investigate the HAPLN1 and GPC5 gene loci for their role in treatment response.

Main Methods:

  • A case-control study involving 199 MS patients treated with interferon-beta for at least two years.
  • Patients were classified as responders or non-responders based on relapse rates and Expanded Disability Status Scale (EDSS) changes.
  • Genotyping of single-nucleotide polymorphisms (SNPs) in the GPC5 and HAPLN1 loci was performed using TaqMan technology.

Main Results:

  • A significant association was found between GPC5 polymorphisms (rs10492503) and interferon-beta response in MS patients (P = 0.0005).
  • No significant association was observed between HAPLN1 polymorphisms and response to interferon-beta therapy (all P > 0.05).

Conclusions:

  • Polymorphisms within the GPC5 gene are confirmed to be associated with interferon-beta treatment response in multiple sclerosis.
  • The HAPLN1 locus did not show a significant association with treatment response in this cohort.
