Purpose of the Study:

  • To investigate the relationship between immediate and late asthmatic responses after repeated allergen inhalations.
  • To determine if prior allergen exposure alters airway reactivity in the late phase.

Main Methods:

  • Sixteen male atopic asthmatics underwent two allergen challenges separated by two weeks.
  • Forced expiratory volume in 1 second (FEV1) was measured for 8 hours post-challenge.
  • Comparison of immediate (within 1 hour) and late (4-8 hours) phase FEV1 changes between challenges.

Main Results:

  • No significant difference in immediate phase FEV1 response between the two challenges.
  • A significantly greater maximal late phase FEV1 decrease on the second challenge (P < 0.01).
  • FEV1 values at 4-8 hours post-challenge were significantly lower on the second day, indicating a more pronounced late bronchial response.

Conclusions:

  • Single allergen challenge increases specific airway reactivity in the late phase upon subsequent rechallenge.
  • This late-phase hyperreactivity appears to persist for at least two weeks after allergen provocation.
  • Findings suggest a mechanism for increased asthma severity with repeated allergen exposure.
