Genome-wide Association Studies-GWAS

Genome-wide association studies or GWAS are used to identify whether common SNPs are associated with certain diseases. Suppose specific SNPs are more frequently observed in individuals with a particular disease than those without the disease. In that case, those SNPs are said to be associated with the disease. Chi-square analysis is performed to check the probability of the allele likely to be associated with the disease.
GWAS does not require the identification of the target gene involved in...

DNA Microarrays

Microarrays are high-throughput and relatively inexpensive assays that can be automated to analyze large quantities of data at a time. They are used in genome-wide studies to compare gene or protein expression under two varied conditions, such as healthy and diseased states. Microarrays consist of glass or silica slides on which probe molecules are covalently attached through surface functionalization. Most commonly, the slides are prepared through the chemisorption of silanes to silica...

Area of Science:

  • Genetics
  • Neuroscience
  • Biotechnology

Background:

  • Single nucleotide polymorphisms (SNPs) are crucial genetic markers for identifying disease susceptibility genes.
  • Complex diseases require high-throughput SNP genotyping methods for large-scale association studies.

Purpose of the Study:

  • To investigate the association between four Brain-Derived Neurotrophic Factor (BDNF) gene polymorphisms and autism.
  • To evaluate a polyacrylamide gel-based microarray with dual-color hybridization as a high-throughput SNP genotyping method.

Main Methods:

  • Applied polyacrylamide gel-based microarray with dual-color hybridization for SNP genotyping.
  • Analyzed four BDNF polymorphisms in autism patients and control groups.
  • Conducted allele, genotype, and haplotype association analyses.

Main Results:

  • The C270T polymorphism in BDNF showed significant differences in allele and genotype frequencies between autism patients and controls.
  • Global haplotype distribution analysis revealed significant differences between the groups.
  • The microarray method proved rapid, simple, and high-throughput for SNP genotyping.

Conclusions:

  • BDNF gene polymorphisms, particularly C270T, may play a role in the pathogenesis of autism.
  • The developed microarray technique is effective for large-scale association studies of genetic susceptibility in complex disorders.
