Factors Affecting Illness

When a person's physical, emotional, intellectual, social development or spiritual functioning is compromised, this deviation from a healthy normal state is called illness. This study investigated the IL-18 rs5744256 polymorphism and physical function in older adults. While not replicated, a meta-analysis suggested a weak association between the C allele and improved walking times in the elderly.

Purpose of the Study:

  • To examine the association between the IL-18 rs5744256 polymorphism and physical functioning in older adults.
  • To replicate or refute prior associations between this genetic variant and locomotor performance.

Main Methods:

  • Analysis of three independent cohorts (N=4,107, aged 60-85) for the IL-18 rs5744256 polymorphism and physical function.
  • Meta-analysis combining new data with existing studies (total N=6,141).
  • Physical function assessed via timed walks and get up and go tests, using reciprocal transformation and z-scores for analysis.

Main Results:

  • The three new cohorts did not show a significant association between the IL-18 rs5744256 polymorphism and physical function.
  • A pooled meta-analysis of all studies revealed weak evidence for an association (regression coefficient = 0.047, 95% CI: 0.010 to 0.083).

Conclusions:

  • The IL-18 rs5744256 polymorphism was not replicated as being associated with physical function in individuals aged 60-85.
  • However, a combined analysis suggests a potential weak beneficial effect of the C allele on walking times in older age.
