    Summary

A new agar gel immunodiffusion test effectively detects antibodies against ovine progressive pneumonia (OPP) virus in sheep. This diagnostic method shows high sensitivity, identifying infected sheep as early as six months post-inoculation.

    Area of Science:

    • Veterinary Immunology
    • Virology
    • Diagnostic Assays

    Background:

    • Ovine progressive pneumonia (OPP) is a significant disease affecting sheep worldwide.
    • Accurate diagnostic tools are crucial for managing OPP outbreaks and herd health.

    Purpose of the Study:

    • To develop and validate an agar gel immunodiffusion (AGID) test for detecting precipitating antibodies against the OPP virus.
    • To assess the sensitivity and specificity of the developed AGID test.

    Main Methods:

    • An AGID test was optimized using purified agar, tris buffer, and sodium chloride.
    • Soluble antigen was prepared from cell cultures persistently infected with OPP virus isolate WLC 1.
    • Serum samples from experimentally inoculated sheep and controls were tested, with results read at 24 and 48 hours.

    Main Results:

    • The AGID test detected precipitating antibodies in sheep experimentally inoculated with OPP virus.
    • Antibody detection varied, with some serums showing one or two lines of precipitate.
    • Sensitivity increased over time, with 25% positivity by month 1, 79% by month 3, and 100% by month 6 post-inoculation.

    Conclusions:

    • The developed AGID test is a viable and adequate method for detecting exposure to the ovine progressive pneumonia virus in sheep.
    • The test's ability to detect antibodies over time supports its use in epidemiological studies and disease management.
