Measuring folate in packed red blood cells (RBCs) offers a practical method for assessing long-term folate status, especially in field studies. This new assay using packed RBCs correlates well with the standard hemolysate method.
Area of Science:
- Clinical Chemistry
- Nutritional Science
- Epidemiology
Background:
- Red blood cell (RBC) folate measurement is preferred for assessing long-term folate status over plasma folate, which reflects recent dietary intake.
- The conventional method for RBC folate analysis requires immediate laboratory processing of whole blood, limiting its use in field studies.
- Packed RBCs can be isolated in field settings, presenting a potential alternative for folate analysis.
Purpose of the Study:
- To validate a new assay for RBC folate analysis using packed RBCs.
- To compare RBC folate values obtained from packed RBCs with those from the routine hemolysate method.
- To assess the practicality of packed RBC folate analysis in field-based epidemiological research.
Main Methods:
- A comparative study involving 50 individuals.
- Folate microbiological assay was used for all measurements.
- RBC folate values from packed RBCs (new assay) were compared to those from the standard hemolysate method (routine assay).
- Plasma folate concentrations were also measured and correlated with both RBC folate assays.
Main Results:
- Both the routine and new RBC folate assays showed statistically significant correlations with plasma folate (r = 0.58 and r = 0.55, respectively; p = 0.001 for both).
- A strong statistically significant correlation was observed between the routine RBC folate assay and the new packed RBC folate assay (r = 0.78, p < 0.001).
Conclusions:
- Measurement of folate in packed RBCs is a practical and validated approach for assessing long-term folate status.
- This method is suitable for field-based and large-scale epidemiological studies where immediate laboratory access for sample processing is not feasible.
- The packed RBC folate assay provides a reliable alternative to the traditional hemolysate method.
