The rare L576P KIT mutation in GISTs leads to aggressive tumors with low imatinib sensitivity. This study confirms L576P/KIT’s poor response to imatinib, even without secondary mutations.

- L576P/KIT showed approximately two-fold reduced sensitivity to imatinib compared to wild-type KIT.
Implications:
- The L576P KIT mutation confers intrinsic resistance to imatinib in GIST, explaining the limited histological response observed.
- These findings highlight the importance of molecular profiling for predicting treatment outcomes in GIST.
- Further research into alternative therapeutic strategies for GIST patients with the L576P mutation is warranted.
