Abstract

Insights

Matrix metalloproteinase-3 (MMP-3) levels are significantly elevated in the ligamentum flavum of degenerative spondylolisthesis patients. This finding suggests MMP-3 may play a key role in the pathogenesis of degenerative spondylolisthesis.

Area of Science:

  • Biochemistry
  • Orthopedics
  • Spinal Surgery

Background:

  • Matrix metalloproteinase-3 (MMP-3) is a key enzyme in extracellular matrix degradation.
  • MMP-3 activity is known in cartilage and intervertebral discs but not previously in ligamentum flavum (LF).

Purpose of the Study:

  • To investigate elevated MMP-3 activity in the LF of patients with degenerative spondylolisthesis (DS).
  • To explore the potential role of MMP-3 in DS pathogenesis.

Main Methods:

  • Human LF samples from 18 DS patients and 18 spinal stenosis (SS) patients were analyzed.
  • Immunohistochemistry, Western blot, and RT-PCR were used to measure MMP-3 expression at protein and mRNA levels.

Main Results:

  • MMP-3 positive cell ratio was significantly higher in LF from DS patients compared to SS patients (P=0.030).
  • Western blot showed higher average optical density of MMP-3 in DS LF (P=0.028).
  • RT-PCR confirmed significantly greater MMP-3 expression in DS patients (P=0.004).

Conclusions:

  • MMP-3 expression in LF is significantly higher in DS patients than in SS patients.
  • Increased MMP-3 expression is associated with degenerative changes in LF and may contribute to DS pathogenesis.
