In-vitro Mutagenesis01:16

In-vitro Mutagenesis

To learn more about the function of a gene, researchers can observe what happens when the gene is inactivated or “knocked out,” by creating genetically engineered knockout animals. Knockout mice have been particularly useful as models for human diseases such as cancer, Parkinson’s disease, and diabetes.
The Eukaryotic Promoter Region02:40

The Eukaryotic Promoter Region

The eukaryotic promoter region is a segment of DNA located upstream of a gene. It contains an RNA polymerase binding site, a transcription start site, and several cis-regulatory sequences.  The proximal promoter region is located in the vicinity of the gene and has cis-regulatory sequences and the core promoter. The core promoter is the binding site for RNA polymerase and is usually located between -35 and +35 nucleotides from the transcription start site. The distal promoter regions are...

Area of Science:

  • Molecular Biology
  • Genetics
  • Developmental Biology

Background:

  • GATA5 is a transcription factor crucial for tissue-specific gene regulation in development.
  • Previous studies showed GATA5 mutations cause embryonic lethality in zebrafish and urogenital defects in mice.

Purpose of the Study:

  • To investigate novel GATA5 gene transcripts and regulatory mechanisms.
  • To understand the functional implications of alternative GATA5 expression.

Main Methods:

  • Identification of alternative promoter and mRNA transcripts using molecular cloning techniques.
  • Reporter gene assays in cultured cells (airway myocytes, bronchial epithelial cells).
  • Analysis of gene expression in transgenic mice.

Main Results:

  • An alternative promoter in intron 1 of the mouse GATA5 gene was identified, generating a 2.5-kb mRNA lacking exon 1.
  • This alternative transcript encodes a truncated "short GATA5" protein (aa 226-404) with a single zinc finger, capable of transactivating gene promoters.
  • A second novel transcript variant was found, differing in the 3'-untranslated region.

Conclusions:

  • The discovery of alternative GATA5 promoters and transcripts expands the known repertoire of GATA5 expression.
  • These findings suggest a more intricate role for GATA5 in regulating gene expression than previously understood.
  • Further research is warranted to elucidate the full functional significance of these GATA5 variants.
