Extraskeletal myxoid chondrosarcoma (ESMC) is a rare tumor. Ultrastructural analysis of its collagen fibrils may aid in diagnosing this soft tissue tumor.

Area of Science:

  • Oncology
  • Pathology
  • Biochemistry

Background:

  • Extraskeletal myxoid chondrosarcoma (ESMC) is a rare soft tissue sarcoma.
  • Accurate diagnosis is crucial for appropriate patient management.

Observation:

  • A case of ESMC in a Japanese woman's thigh was examined.
  • Tumor cells exhibited chondrocyte-like features and S100 protein positivity.
  • Advanced electron microscopy techniques were employed.

Findings:

  • Quick-freezing and deep-etching (QF-DE) revealed unique collagen fibril ultrastructure in the myxoid matrix.
  • Collagen fibrils were thin (20-35 nm) with pleated surfaces and obscure 65 nm striations.
  • These ultrastructural features are characteristic of the ESMC myxoid matrix.

Implications:

  • The QF-DE method highlights specific collagen fibril characteristics in ESMC.
  • This ultrastructural finding aids in the differential diagnosis of myxoid soft tissue tumors.
  • Further research into ESMC matrix composition could improve diagnostic accuracy.
