Liver Regeneration01:24

Liver Regeneration

The liver is an important organ in vertebrates that plays an essential role in metabolism. It is also responsible for storing and redistributing nutrients such as carbohydrates, fats, and vitamins in the body. Additionally, the liver releases bile salts which are critical for digesting food and eliminating toxic metabolites from the body.
Cells of Liver
The liver comprises four major types of cells— hepatocytes, stellate, Kupffer, and sinusoidal endothelial cells. The hepatocytes are large...

Summary
This summary is machine-generated.

Complement activation and C5a-receptor changes occur after liver resection (LR). Polymorphonuclear cells (PMN) show altered C5a-dependent responses, impacting liver regeneration post-surgery.

Area of Science:

  • Immunology
  • Hepatology
  • Surgical Research

Background:

  • Complement activation plays a role in regulating liver regeneration after liver resection (LR).
  • Understanding the role of complement system components and their receptors in clinical LR is crucial.

Purpose of the Study:

  • To investigate the hypothesis that complement activation and changes in C5a-receptors (C5aR, C5L2) on polymorphonuclear cells (PMN) and monocytes are important in clinical LR.
  • To assess the functional responses of PMN and monocytes following LR.

Main Methods:

  • Plasma levels of anaphylatoxins (C3a, C4a, C5a) and C5b9 were measured in 25 patients undergoing LR.
  • Receptor expression (C5aR, C5L2) on PMN and monocytes was assessed using flow cytometry (FACS) in 19 patients.
  • In vitro PMN functional assays, including chemotaxis, L-selectin shedding, and Mac-1 expression, were performed.

Main Results:

  • Complement components C3a and C5b9 increased significantly after LR, while C4a and C5a decreased.
  • C5L2 expression on PMN decreased at 4 hours post-LR and increased at 24 hours.
  • PMN exhibited diminished C5a-dependent chemotactic response and reduced activation markers (L-selectin shedding, Mac-1 expression) post-LR. Monocyte C5L2 expression changes correlated with impaired liver function.

Conclusions:

  • Complement components are released into circulation following clinical liver resection.
  • These changes lead to altered C5a-dependent functional responses in polymorphonuclear cells (PMN).
  • The study highlights the involvement of the complement system in the post-operative recovery phase after LR.
