Genetic variations in APOE influence renal cell carcinoma (RCC) risk. Specific single nucleotide polymorphisms (SNPs) within the APOE promoter region were linked to increased susceptibility to this cancer. This suggests a role for lipid peroxidation pathways in RCC development.
Area of Science:
- Genetics
- Oncology
- Biochemistry
Background:
- Lipid peroxidation is a key mechanism linking risk factors to renal cell carcinoma (RCC).
- Genetic variations may modify an individual's susceptibility to RCC through pathways like lipid peroxidation.
Purpose of the Study:
- To investigate if genes involved in lipid peroxidation influence renal cell carcinoma (RCC) risk.
- To identify specific genetic variants associated with RCC susceptibility.
Main Methods:
- Genotyped 635 single nucleotide polymorphisms (SNPs) in 38 candidate genes in European and US case-control studies.
- Replicated top candidate SNPs in a second US study.
- Analyzed association of SNPs and haplotypes with RCC risk.
Main Results:
- Two SNPs (rs8106822 and rs405509) in the APOE promoter region replicated in both studies.
- The rs8106822 variant showed increased RCC risk (P(trend) = 0.001 combined).
- The rs405509 variant and a G-G haplotype showed decreased RCC risk (P(trend) = 0.001 combined).
- rs405509 demonstrated in vitro effects on APOE transcriptional activity.
Conclusions:
- Findings identify a functional region at the APOE locus associated with RCC susceptibility.
- Genetic variation in APOE may play a role in renal cell carcinoma development.
- These results contribute to understanding the genetic basis of RCC.

Comparing Copy Number Variations and SNPs
Copy number variations or CNVs are the structural variations that cover more than 1kb of DNA sequence. The single nucleotide polymorphism (SNP), on the other hand, is a single nucleotide change or a point mutation that is found in more than 1%...
