Area of Science:

  • Molecular Biology
  • Neuroscience
  • Genomics

Background:

  • Microdissection enables transcriptome analysis of specific cell populations from complex tissues.
  • RNA instability during immunolabeling hinders microdissection for genomic studies.

Purpose of the Study:

  • To develop and validate a novel immunolabeling method for preserving RNA integrity during cell microdissection.
  • To enable high-quality transcriptome analysis from isolated cells of heterogeneous tissues.

Main Methods:

  • Developed an immunolabeling protocol using a high salt buffer to stabilize RNA.
  • Assessed RNA integrity using three methods after incubation in high salt buffer.
  • Validated immunolabeling compatibility and performed laser microdissection on immunolabeled dopamine neurons.

Main Results:

  • High salt buffer incubation (≥20 h) maintained RNA integrity comparable to fresh-frozen tissue.
  • RNA integrity was superior to rapid immunolabeling procedures.
  • Successful immunolabeling and laser microdissection of dopamine neurons with preserved RNA integrity.

Conclusions:

  • The high salt buffer immunolabeling method effectively stabilizes RNA for microdissection.
  • This technique is crucial for accurate transcriptome analysis of specific cell types from complex tissues.
  • Enables advanced genomics investigations of normal and pathological cells.
