Background:

  • Apolipoprotein D (apo D) is a lipocalin found in the nervous system, implicated in reinnervation, regeneration, and neuronal protection.
  • Altered apo D expression is linked to normal brain aging and neurodegenerative diseases in specific brain regions.

Purpose of the Study:

  • To investigate the regional and cellular expression patterns of apo D in the normal human brain throughout the aging process.
  • To understand the role of apo D in central nervous system aging and its potential protective mechanisms.

Main Methods:

  • Utilized slot-blot techniques for quantifying protein and mRNA levels of apo D.
  • Employed immunohistochemistry and hybridohistochemistry for cellular localization of apo D synthesis.
  • Examined post-mortem human cerebral cortex samples.

Main Results:

  • A positive correlation was observed between apo D expression and chronological aging.
  • Both apo D mRNA and protein levels were significantly higher in white matter compared to grey matter.
  • Immunohistochemistry and in situ hybridization revealed apo D synthesis in both neurons and glial cells, notably oligodendrocytes. The number of apo D-synthesizing neurons increased with age.

Conclusions:

  • Apolipoprotein D plays a fundamental role in the aging of the central nervous system.
  • Increased apo D expression with age may represent a cellular defense mechanism against lipid peroxidation products and other age-related deleterious effects.
