Confocal Fluorescence Microscopy

Confocal microscopy is an advanced microscopic technique. The prime advantage of the confocal microscope over other microscopy techniques is its ability to block the out-of-focus light from the illuminated samples using pinholes. Background:

  • Schistosoma mansoni infection leads to reduced egg output in hosts during chronic phases.
  • Ageing is a potential factor contributing to decreased reproductive capacity in schistosomes.

Purpose of the Study:

  • To investigate the effects of ageing on the morphology of adult Schistosoma mansoni worms.
  • To understand the mechanisms behind age-related infertility in schistosomes.

Main Methods:

  • Confocal laser scanning microscopy was used to examine adult Schistosoma mansoni worms.
  • Worms were recovered from experimentally infected Nectomys squamipes (water rat) after prolonged infection periods.

Main Results:

  • While some reproductive organs (ovary, vitellaria, oocytes) appeared normal, the ootype showed signs of impaired eggshell production.
  • Male worms exhibited empty areas in testicular lobes and flaccid dorsal musculature.
  • Infected rats passed viable eggs for 65 weeks, followed by unviable eggs until death at 70 weeks.

Conclusions:

  • Older male and female Schistosoma mansoni worms display morphological changes indicative of infertility.
  • Ageing significantly impacts the reproductive capabilities of schistosomes, affecting egg viability and production.
