CD2-associated protein (CD2AP) is crucial for Natural Killer (NK) cell cytotoxicity. Reduced CD2AP impairs NK cell conjugate formation and granule transport, significantly hindering cancer cell killing.
Area of Science:
- Immunology
- Cell Biology
Background:
- CD2-associated protein (CD2AP) is a cytoplasmic protein involved in cellular processes like actin remodeling and membrane trafficking.
- CD2AP's role in T cell signaling and structure is established, but its function in Natural Killer (NK) cell-mediated cytotoxicity was unknown.
Purpose of the Study:
- To investigate the role of CD2AP in the cytotoxic function of NK cells.
- To determine the molecular mechanisms by which CD2AP might influence NK cell killing.
Main Methods:
- Utilized CD2AP knockdown in the human NK92 cell line.
- Assessed cytotoxicity, conjugate formation, and degranulation.
- Performed co-localization studies and immunoprecipitation to examine CD2AP-FasL interactions.
Main Results:
- CD2AP knockdown significantly reduced NK92 cell cytotoxicity, irrespective of target cell FasL sensitivity.
- Impaired conjugate formation and degranulation were observed in CD2AP-deficient NK92 cells.
- CD2AP was found to associate with FasL and facilitate the polarization of FasL-bearing granules toward the target cell contact site.
Conclusions:
- CD2AP plays a critical role in NK cell-mediated killing by promoting conjugate formation and directed transport of cytotoxic granules.
- CD2AP's interaction with FasL is important for efficient NK cell cytotoxicity.
