Background:

  • Nephrectomized rats are models for chronic renal failure.
  • Renal cell carcinoma (RCC) was observed in remnant kidneys post-uninephrectomy (UNX).

Purpose of the Study:

  • To investigate the insulin-like growth factor (IGF)-1 signaling pathway in UNX-induced renal cancer.
  • To characterize a novel rat model of kidney cancer.

Main Methods:

  • Adult male Sprague-Dawley rats underwent UNX or sham surgery.
  • Tumor tissues were analyzed at 3, 7, and 10 months post-surgery.
  • Western blot was used to assess protein expression.

Main Results:

  • UNX rats developed renal cell carcinoma by 7 months post-surgery, with 68.2% exhibiting invasive carcinoma.
  • Increased expression of proliferating cell nuclear antigen was observed.
  • Down-regulation of IGF binding protein 3 and up-regulation of protein kinase Czeta and Akt/protein kinase B were detected.

Conclusions:

  • Uninephrectomy in rats provides a unique model for studying renal cancer.
  • The IGF-1 signaling pathway is enhanced in UNX-induced renal cancer.
  • This model facilitates research into kidney cancer mechanisms and therapeutic targets.
