Area of Science:

  • Molecular Biology
  • Biotechnology
  • Histopathology

Background:

  • Formalin-fixed, paraffin-embedded (FFPE) tissues are crucial for molecular analysis.
  • Formalin fixation causes cross-linking, complicating RNA extraction and leading to variable quality.
  • Existing RNA extraction methods for FFPE tissues often yield inconsistent results.

Purpose of the Study:

  • To develop a reliable RNA extraction method for FFPE tissues.
  • To improve RNA quality and yield for downstream molecular profiling.
  • To enhance the utility of archival FFPE samples in research.

Main Methods:

  • Optimized deparaffinization at high temperatures.
  • Extended 3-day lysis period at 65°C.
  • Utilized bioanalyzer for RNA fragment length analysis.

Main Results:

  • Achieved an average RNA yield of 4.5-5.5 ng/mm³ from archival FFPE tissue.
  • Obtained 260/280 ratios between 1.80 and 1.95, indicating good RNA purity.
  • Successfully amplified 300 bp amplicons via reverse transcription and PCR, despite shorter RNA fragment lengths.

Conclusions:

  • The optimized method provides reliable RNA extraction from FFPE tissues.
  • Improved RNA yield and quality facilitate molecular profiling studies.
  • This technique increases the value of FFPE tissues for research and diagnostics.
