This study developed a method to measure oxidative DNA damage in human trabecular meshwork from glaucoma patients. The findings suggest a role for oxidative stress in glaucoma pathogenesis.
Area of Science:
- Ophthalmology
- Molecular Biology
- Genetics
Background:
- Oxidative DNA damage is implicated in degenerative eye diseases like glaucoma.
- Evaluating this damage directly in human ocular tissues is crucial for understanding disease mechanisms.
Purpose of the Study:
- To develop and validate a method for assessing oxidative DNA damage in human trabecular meshwork (HTM) from glaucoma patients.
- To simultaneously analyze genetic variations in antioxidant defense genes (GSTM1 and GSTT1) within the same DNA samples.
Main Methods:
- DNA extraction from HTM samples obtained during surgery from glaucoma patients and controls.
- Quantification of oxidative DNA damage marker 8-hydroxy-2'-deoxyguanosine (8-OHdG) using 32P postlabeling and electronic autoradiography.
- Genotyping of GSTM1 and GSTT1 polymorphisms via polymerase chain reaction (PCR) and agarose electrophoresis.
Main Results:
- The developed method successfully quantified 8-OHdG in minimal DNA amounts (0.11 µg) from all HTM samples.
- Elevated levels of 8-OHdG were detected in glaucoma patients compared to controls, indicating increased oxidative DNA damage.
- GSTM1 and GSTT1 genotypes were accurately determined from the same DNA extracts, demonstrating method efficiency.
Conclusions:
- This novel method enables direct evaluation of oxidative DNA damage in human trabecular meshwork from glaucoma patients.
- The findings support the hypothesis that oxidative stress contributes to the pathogenesis of glaucoma.
- The technique's efficiency with small sample volumes facilitates research using surgically obtained ocular tissues.
