Ingenuity pathway analysis of clozapine-induced obesity

Background:
Lipid accretion is one of the major side effects of clozapine pharmacotherapy of schizophrenia that made clozapine into an interesting obesity drug model.
Method:
Ingenuity Pathway Analysis (IPA) engine was used for core analysis and building the networks of weight regulation.
Results:
The examination of molecules that were selected into 'clozapine neighborhood' identified them as multifunctional signals that appear to orchestrate vascular and tissue functions plausibly implicated in adiposity side effect.
Conclusions:
It is hypothesized that clozapine unmasks the functional and morphological phenotype of microvascular deficit that facilitates shunting nutrients from utilization toward storage.
Insights
Clozapine, used for schizophrenia, may model obesity by affecting nutrient storage. This drug may unmask microvascular deficits, leading to increased fat accumulation and weight gain.
Area of Science:
- Pharmacology
- Metabolic Research
- Schizophrenia Treatment
Background:
- Clozapine is a key antipsychotic for schizophrenia treatment.
- Lipid accretion is a significant side effect of clozapine therapy.
- This side effect makes clozapine a model for studying obesity.
Purpose of the Study:
- To investigate the molecular mechanisms behind clozapine-induced weight gain.
- To understand how clozapine influences lipid accretion and obesity.
Main Methods:
- Ingenuity Pathway Analysis (IPA) was employed for core analysis.
- Network analysis was used to map weight regulation pathways.
Main Results:
- Molecules in the 'clozapine neighborhood' are multifunctional signals.
- These signals appear to regulate vascular and tissue functions.
- These functions are plausibly linked to the adiposity side effect.
Conclusions:
- Clozapine may reveal a microvascular deficit phenotype.
- This deficit could facilitate nutrient shunting towards storage.
- This mechanism contributes to clozapine-associated obesity.
