α-Hydroxy Ketones via Reductive Coupling of Esters: Acyloin Condensation Overview01:19

α-Hydroxy Ketones via Reductive Coupling of Esters: Acyloin Condensation Overview

The pinacol and McMurry reactions involve the reductive coupling of ketones or aldehydes. Similarly, the bimolecular reductive coupling of two ester molecules in the presence of sodium metal in an aprotic solvent yields an α-hydroxy ketone product. The α-hydroxy ketone is also called acyloin, so the reaction is referred to as ‘acyloin condensation.’
Alkylation of β-Ketoester Enolates: Acetoacetic Ester Synthesis01:07

Alkylation of β-Ketoester Enolates: Acetoacetic Ester Synthesis

Acetoacetic ester synthesis is a method to obtain ketones from alkyl halides and β-keto esters. The reaction occurs in the presence of an alkoxide base that abstracts the acidic proton of the β-keto esters. The step results in an enolate ion which is doubly stabilized. The enolate then reacts with an alkyl halide via the SN2 process to produce an alkylated ester intermediate with a new C–C bond. The hydrolysis of the intermediate, followed by acidification, results in an alkylated β-keto acid.

Acetals and Thioacetals as Protecting Groups for Aldehydes and Ketones01:24

Acetals and Thioacetals as Protecting Groups for Aldehydes and Ketones

Acetals are formed by reacting two equivalents of alcohol with carbonyl compounds like aldehydes or ketones. Acetals are unaffected by bases, nucleophiles, oxidizing agents, and reducing agents. They serve as protecting groups for aldehydes and ketones. Acetals can be easily formed and also easily removed via mild acid hydrolysis.
In the presence of multiple functional groups, when selective reduction of one group over the other is desired, groups like aldehydes and ketones that form acetals...

Alkylation of β-Diester Enolates: Malonic Ester Synthesis

Malonic ester synthesis is a method to obtain α substituted carboxylic acids from ꞵ-diesters such as diethyl malonate and alkyl halides.

Summary
This summary is machine-generated.

Cockroach allergen Per a 3, a potent asthma trigger, has been structurally characterized for the first time. Its hexameric form and accessible epitopes explain its high allergic potential and stability.

Area of Science:

  • Allergen research
  • Structural biology
  • Immunology

Background:

  • Cockroach allergens, particularly Per a 3, are significant global asthma triggers.
  • Previous research lacked biochemical and biophysical data on Per a 3 family members.
  • Recent studies link Per a 3 to increased allergy indicators in Europeans.

Purpose of the Study:

  • To perform a thorough structural characterization of cockroach Per a 3 allergens.
  • To investigate the biochemical and biophysical properties of Per a 3 isoforms.
  • To understand the structural basis of Per a 3 allergenicity and stability.

Main Methods:

  • Isolation and characterization of two Per a 3 isoforms (P I and P II).
  • Determination of molecular masses and sedimentation coefficients.
  • Sequence analysis to identify subunit types.
  • Homology modeling to generate a 3D model of the hexameric allergen.

Main Results:

  • Two Per a 3 isoforms (P I, P II) isolated as hexamers with distinct molecular masses.
  • Sedimentation coefficients determined: 17.4+/-0.7 S (P II) and 19.0+/-0.9 S (P I).
  • Identified known (Per a 3.01, Per a 3.0201) and a novel (Per a 3.03) allergenic subunits.
  • 3D model revealed accessible antigenic peptides and IgE-epitopes on the hexamer surface.

Conclusions:

  • The hexameric structure of Per a 3 with surface-exposed epitopes likely contributes to its high allergic potential.
  • The structural characteristics may also explain the extreme stability of Per a 3.
  • This study provides crucial insights into the molecular mechanisms of cockroach allergy.
