Super-resolution Fluorescence Microscopy

Super-resolution fluorescence microscopy (SRFM) provides a better resolution than conventional fluorescence microscopy by reducing the point spread function (PSF). PSF is the light intensity distribution from a point that causes it to appear blurred. Due to PSF, each fluorescing point appears bigger than its actual size, and it is the PSF interference of nearby fluorophores that causes the blurred image. Various approaches to achieving higher resolution through SRFM have recently been developed.

Background:

Picosecond time-resolved transmission Raman spectroscopy is a valuable technique for probing material properties.
Understanding photon migration is crucial for interpreting spectroscopic data from scattering media.

Purpose of the Study:

To model photon migration in trans-stilbene powder using picosecond time-resolved transmission Raman spectroscopy.
To investigate the spatial resolution of transmission Raman spectroscopy in the diffusion regime.

Main Methods:

Acquisition of picosecond time-resolved transmission Raman data from 1 mm thick trans-stilbene powder samples.
Development and application of a Monte Carlo model to simulate laser and Raman pulse profiles and photon migration.
Analysis of photon flight paths and spatial resolution (lateral and depth) as a function of experimental parameters.

Main Results:

Photon migration broadened the incident probe pulse by two orders of magnitude.
Transmitted Raman pulses were broader and propagated slower than transmitted laser pulses.
Lateral spatial resolution was found to worsen linearly with sample thickness, approximately 50% of the sample thickness.
Time-resolved experiments show potential for improved lateral resolution by detecting 'snake' photons.

Conclusions:

Monte Carlo modeling accurately simulates photon migration effects in transmission Raman spectroscopy.
Spatial resolution in transmission Raman spectroscopy is fundamentally limited by photon diffusion in thick samples.
Time-resolved techniques offer a pathway to enhance spatial resolution in transmission Raman spectroscopy.
