Glycosaminoglycans

Glycosaminoglycans (GAGs), also known as mucopolysaccharides, are long and linear polymers comprising of specific repeating disaccharides - the amino sugar that can be N-acetylglucosamine or N-acetylgalactosamine, and a uronic acid that is usually glucuronic acid or iduronic acid.
GAGS are found in the extracellular matrix of vertebrates, invertebrates, and bacteria. Due to their polar nature they attract water, and serve as excellent lubricants or shock absorbers in an animal body.

A new microassay accurately measures alpha-mannosidase in synovial fluid. Enzyme levels correlate with inflammation and cell counts in joint disease, offering a quantitative diagnostic tool.

Area of Science:

  • Biochemistry
  • Rheumatology
  • Enzymology

Background:

  • Synovial fluid analysis is crucial for diagnosing joint diseases.
  • Quantifying inflammatory markers in synovial fluid aids in disease assessment.
  • Lysosomal glycosidases, like alpha-mannosidase, are implicated in inflammatory processes.

Purpose of the Study:

  • To develop and validate a microassay for synovial fluid alpha-mannosidase activity.
  • To investigate the correlation between alpha-mannosidase activity and indicators of joint inflammation.
  • To explore the potential of alpha-mannosidase as a biomarker for arthropathic conditions.

Main Methods:

  • Development of a microassay technique for alpha-mannosidase activity measurement.
  • Analysis of 184 synovial fluid samples from patients with diverse arthropathic conditions.

  • Correlation analysis between enzyme activity, synovial fluid cell count, and inflammation markers.
  • Main Results:

    • The microassay demonstrated reliable measurement of synovial fluid alpha-mannosidase.
    • A significant correlation was observed between enzyme activity and synovial fluid cell count.
    • Enzyme activity closely correlated with the degree of inflammation in various joint diseases.

    Conclusions:

    • Synovial fluid alpha-mannosidase activity can be reliably quantified using the developed microassay.
    • Alpha-mannosidase activity serves as a quantitative indicator of inflammation in joint diseases.
    • Evaluation of alpha-mannosidase and other lysosomal glycosidases offers a valuable method for assessing inflammatory joint disease.
