Background:

  • Subcutaneous Immunotherapy (SCIT) is a primary treatment for insect venom allergy, modifying allergic responses.
  • The efficacy of SCIT in insect venom allergy is well-established.
  • Basophil activation, measured by CD203c and CD63 expression, is a potential indicator of treatment response.

Purpose of the Study:

  • To investigate the change in basophil sensitivity during the up-dosing phase of SCIT for wasp allergy.
  • To compare CD203c and CD63 as markers for basophil activation.
  • To assess the protective effect of soluble factors in patients undergoing SCIT.

Main Methods:

  • 18 patients undergoing SCIT for wasp allergy were enrolled.
  • Blood samples were collected at various time points during SCIT up-dosing.
  • Basophil activation was measured using flow cytometry stimulated with allergen, analyzing CD203c and CD63 expression.

Main Results:

  • Basophil activation markers CD63 and CD203c showed similar results.
  • Significant reduction in basophil sensitization was observed three weeks after SCIT initiation.
  • An increase in the protective plasma component was noted upon reaching maintenance allergen concentrations.
