Summary

Fascin protein is expressed in about one-third of colorectal cancers and is linked to lymph node metastasis and advanced tumor stage. Its presence may indicate a poorer prognosis in later-stage colorectal carcinoma.

Background:

  • Understanding molecular markers associated with colorectal cancer progression is crucial for improved diagnostics and therapeutics.

Purpose of the Study:

  • To investigate the clinical significance of fascin protein expression in colorectal carcinoma.
  • To determine the association between fascin expression and clinicopathological features and patient survival.

Main Methods:

  • A retrospective analysis of 167 colorectal adenocarcinoma cases.
  • Immunohistochemistry was used to assess fascin expression and quantify it using an immunohistochemical score (IHS).
  • Statistical analysis correlated fascin expression with lymph node metastasis, tumor stage, and survival rates.

Main Results:

  • Fascin was expressed in 35.3% of colorectal carcinomas, with higher expression linked to lymph node metastasis (p=0.002) and advanced tumor stage (p=0.007).
  • A strong correlation was observed between fascin expression at invasive borders and lymph node metastasis (r=0.747, p=0.005).
  • In advanced stages (III/IV), patients with high fascin expression (IHS>10) had significantly lower two-year survival rates (60.0%) compared to those without (92.2%, p=0.003).

Conclusions:

  • Fascin is heterogeneously expressed in colorectal carcinoma and is significantly associated with lymph node metastasis and advanced tumor stage.
  • Fascin expression may serve as a prognostic marker for colorectal cancer, potentially indicating a role in lymph node metastasis.
