Summary

Major depressive disorder (MDD) in elderly women did not show significant brain structural changes. Aging, not depression, was linked to alterations in cortical thickness and gray matter density.

Background:

  • Major depressive disorder (MDD) is prevalent in the elderly.
  • Previous research suggests links between depression and neurobiological abnormalities, but findings in older adults are inconsistent.
  • The relationship between lifetime depression and brain structure in older age requires further investigation.

Purpose of the Study:

  • To concurrently examine cortical thickness and voxel-based morphometric (VBM) abnormalities in elderly female patients with MDD.
  • To determine if previous or current depression is associated with neurobiological abnormalities in older age.
  • To explore the impact of depression severity and episode history on brain structure.

Main Methods:

  • Magnetic resonance imaging (MRI) was used to collect data from 28 female elderly subjects with MDD and 38 age-matched controls.
  • Cortical thickness measurements and VBM analysis were applied to the MRI data.
  • Statistical analyses were performed to compare brain structure between groups and correlate with depression metrics.

Main Results:

  • No significant differences in cortical thickness or gray matter density were found between elderly women with MDD and controls.
  • Lifetime depression, illness severity, and number of depressive episodes were not associated with neurobiological abnormalities.
  • Significant age-related effects on cortical thickness and VBM were observed across all participants, irrespective of diagnosis.

Conclusions:

  • In contrast to some literature, this study suggests that lifetime and current depression do not significantly influence brain structure in elderly females.
  • Age-related changes in brain structure are prominent in this population, independent of major depressive disorder.
  • Further research is needed to understand the complex interplay between aging, depression, and brain health.
