We developed a robust method for isolating high-quality RNA from individual zebrafish embryos. This single-embryo approach eliminates the need for pooling, preserving biological variation for more accurate transcriptome analysis.

Area of Science:

  • Developmental Biology
  • Molecular Biology
  • Genomics

Background:

  • Traditional transcriptome analysis requires pooling embryos, confounding individual biological variation.
  • Existing methods are not feasible for single zebrafish embryo RNA isolation.

Purpose of the Study:

  • To develop a robust method for isolating abundant, high-quality RNA from individual zebrafish embryos.
  • To enable single-embryo transcriptome analysis, preserving biological variation.

Main Methods:

  • Homogenization of single embryos in liquid nitrogen.
  • RNA extraction using phenol and column purification.
  • Method validated across all zebrafish embryonic stages.

Main Results:

Isolation and Characterization of Single Cells from Zebrafish Embryos

Published on: March 12, 2016

  • High-quality RNA obtained (average RIN 8.3-8.9).
  • Yield consistently >/= 200 ng RNA per embryo.
  • High success rate (~100%) with low technical variation in downstream microarray experiments.
  • Conclusions:

    • A high-quality, robust RNA isolation method for single embryos is presented.
    • Eliminates sample pooling, preserving biological variation.
    • Applicable to zebrafish and other model systems for various applications (e.g., (q)PCR, sequencing).
