    Background:

    • Adrenocorticotropin (ACTH) secretion is crucial for stress response.
    • Negative-feedback mechanisms regulate ACTH release.
    • The kinetics of feedback regulation are not fully understood.

    Purpose of the Study:

    • To quantitatively analyze the fast, rate-sensitive negative-feedback regulation of ACTH secretion during stress.
    • To determine the critical rate of plasma corticosterone rise for feedback inhibition.

    Main Methods:

    • Rats were anesthetized with pentobarbital sodium.
    • Plasma corticosterone levels were manipulated via infusion.
    • Histamine was used as a stressor to elicit ACTH secretion.
    • Serial blood sampling from the carotid artery was performed.

    Main Results:

    • A fast, rate-sensitive feedback inhibition of ACTH secretion was observed when plasma corticosterone was rising.
    • The inhibition was independent of the absolute corticosterone level.
