Insights

Sunitinib showed clinical activity in previously treated advanced non-small cell lung cancer (NSCLC) in Asian patients. This oral targeted therapy was generally well-tolerated, suggesting potential for this patient group.

Area of Science:

  • Oncology
  • Pharmacology

Background:

  • Sunitinib is an approved oral multitargeted tyrosine kinase inhibitor for advanced renal cell carcinoma and gastrointestinal stromal tumors.
  • Limited research exists on sunitinib's efficacy in non-small cell lung cancer (NSCLC), particularly in Asian populations.

Purpose of the Study:

  • To evaluate the clinical activity and tolerability of sunitinib in Asian patients with previously treated advanced NSCLC.

Main Methods:

  • An open-label, non-randomized phase II trial was conducted.
  • 22 Asian patients with stage IIIB or IV NSCLC, who progressed on platinum-based chemotherapy, were enrolled.

Main Results:

  • The objective response rate (ORR) was 9% (2 partial responses).
  • An additional 7 patients achieved stable disease (SD) for at least 8 weeks.
  • Median progression-free survival (PFS) was 11.8 weeks, and median overall survival (OS) was 23.4 weeks.
  • Treatment was generally well-tolerated with a favorable toxicity profile.

Conclusions:

  • Sunitinib demonstrates substantial single-agent activity and a favorable toxicity profile in pretreated advanced NSCLC Asian patients.
  • Further large-scale, high-quality prospective trials are warranted to confirm these findings.
