Summary

A new bioluminescence imaging method allows in vivo measurement of cytotoxic T lymphocyte (CTL) function in the liver. This technique accurately quantifies CTL antiviral activity, improving understanding of hepatic immune regulation.

Background:

  • Assessing cytotoxic T lymphocyte (CTL) function in the liver in vivo is challenging.
  • Current methods like alanine aminotransferase levels lack sensitivity for detecting T cell immunity against low-level hepatocyte infection.

Purpose of the Study:

  • To develop a sensitive in vivo method for evaluating CTL effector function in the liver.
  • To enable simultaneous detection of viral infection and quantification of antiviral CTL responses.

Main Methods:

  • Adenoviral transfer of reporter and immune target genes into hepatocytes.
  • Bioluminescence imaging to detect reporter gene expression.
  • In vivo quantification of antiviral CTL activity.

Main Results:

  • Bioluminescence imaging detected as few as 10,000 infected hepatocytes.
  • The method quantified antiviral effector function of as few as 50,000 CTLs.
