Purpose of the Study:

  • To investigate the individual and combined effects of female sex and behaviorally inhibited temperament on the acquisition of active-avoidance responses in an animal model.
  • To determine if genetic factors (strain and sex) influence the development and persistence of anxiety-related behaviors.

Main Methods:

  • Utilized a discrete trial lever-press escape-avoidance protocol in Sprague-Dawley (SD) and Wistar-Kyoto (WKY) rats.
  • Assessed the acquisition of behavioral avoidance, reinforced responses, and non-reinforced responses in male and female rats.
  • Compared behavioral differences between WKY rats (genetically predisposed to inhibition) and SD rats, and between sexes within each strain.

Main Results:

  • WKY rats, regardless of sex, acquired reinforced avoidance responses faster than SD rats and emitted more non-reinforced responses.
  • SD females acquired reinforced responses quicker and emitted more non-reinforced responses than SD males, indicating sex differences in this strain.
  • Both behavioral inhibition (WKY strain) and female sex were associated with increased avoidant behavior, but their effects were not additive.

Conclusions:

  • Genetics, specifically strain and sex, plays a significant role in anxiety vulnerability and the acquisition of active-avoidance responses.
  • Behavioral inhibition and female sex independently contribute to avoidant behaviors, but do not synergistically increase vulnerability.
  • This study underscores the importance of considering genetic background in animal models of anxiety and avoidance learning.
