Summary

Guanosine reduces cancer stem cell populations by affecting transporter activity. This study shows guanosine impacts side population cells and ABCG2 transporter expression in lung cancer, though other transporters also influence the side population phenotype.

Background:

  • Cancer stem cells (CSCs) reside in the 'side population' (SP) and drive tumor growth.
  • ATP-binding cassette transporter G2 (ABCG2) is strongly associated with the SP phenotype.
  • Guanosine, a signaling molecule, influences stem cell proliferation and cholesterol efflux via ABCG transporters.

Purpose of the Study:

  • To investigate the effect of guanosine on the SP of A549 lung cancer cells.
  • To determine if guanosine influences ABCG2 expression in A549 lung cancer cells.

Main Methods:

  • Fluorescence-activated cell sorting (FACS) to quantify SP cells.
  • Western blot analysis to assess ABCG2 transporter expression.

Main Results:

  • Guanosine (10 microM) significantly decreased the proportion of SP cells after 48 hours.
  • Guanosine (10 microM) significantly decreased ABCG2 expression after 6 hours.
  • The observed effects on SP cells and ABCG2 expression showed a temporal dissociation.

Conclusions:

  • Guanosine influences both the SP cell proportion and ABCG2 transporter levels in A549 lung cancer cells.
  • The lack of direct correlation between ABCG2 expression and SP phenotype suggests other transporters are involved in maintaining the SP in these cells.
