Background:

  • Emerging reports suggest a potential link between benfluorex and cardiac valve regurgitation.

Purpose of the Study:

  • To investigate the association between benfluorex use and unexplained mitral regurgitation.
  • To quantify the risk of mitral regurgitation in patients exposed to benfluorex.

Main Methods:

  • A case-control study was conducted between 2003 and 2009.
  • Cases were patients with unexplained mitral regurgitation; controls had explained mitral regurgitation.
  • Drug exposure, including benfluorex, was assessed blindly.

Main Results:

  • Out of 682 eligible patients, 27 cases and 54 controls were identified.
  • Benfluorex use was reported in 19 of 27 cases versus 3 of 54 controls.
  • An odds ratio of 17.1 (95% CI: 3.5-83) was found for benfluorex use and unexplained mitral regurgitation.

Conclusions:

  • Benfluorex use is significantly associated with unexplained mitral regurgitation.
  • The findings highlight a potential safety concern for benfluorex regarding cardiac health.
